The Sutherland Sharks product was released at the end of his contract with English Premier League club Tottenham Hotspur in June.
The 21-year-old Olyroos goalkeeper featured for Spurs in their under-18s, 21s and 23s sides but did not make a first team appearance.
Glover has already played in the A-League, featuring in a season-long loan with the Central Coast for the 2017-18 campaign but only made four appearances. He also spent time on loan in the Swedish first division at Helsingborg.
Glover, from Bangor, made nine appearances for Australia's under-20s and has three caps for the under-23s.
"I'm excited to join up with Melbourne City and to work with [head coach] Erick [Mombaerts] and [goalkeeper coach] Neil [Young]," Glover said.
"I am committed to working hard and learning the new style of play Erick is implementing at Melbourne City, developing [my] ball-playing skills and game-management.
"I want to challenge myself and compete for a regular spot in the side and look forward to getting stuck in."
Melbourne City director of football Michael Petrillo said Glover was one of Australia's brightest goalkeeping talents.
"We're looking forward to working with him to help him continue his impressive development and challenge for game time this season," Petrillo said.
"Tom has been in elite set ups before during his time with the national side and in the UK with Tottenham Hotspur and will be able to share those high standards with other young players at the club."
Glover will look to establish himself as City's No.1 goalkeeper, with Dean Bouzanis the club's only other senior shot stopper.
