Health officials say nearly 200 people were tested in the first two days after the low-key opening of the drive-through service in the Multi-Purpose Centre in Flora Street on Tuesday.
The new service adds to a growing range of testing options.
People who are feeling unwell can also make a phone or video appointment with a GP, who makes an assessment and safely arranges a test if needed.
In addition, privately run testing clinics have recently opened, joining those operated at Sutherland Hospital and St George Hospital by South Eastern Sydney Local Health District.
Dr Marianne Gale, the health district's director, population and community health, said anyone experiencing fever or respiratory symptoms - even if mild - was encouraged to promptly seek testing.
"I would urge anyone experiencing even mild symptoms like a scratchy throat or runny nose to be tested," she said.
Dr Michael Moore, chief executive of Central and Eastern Sydney Primary Health Network, said people could not only talk to a GP about coronavirus but also should continue to seek general health care.
"Your GP is still there for you, so please do not put off or miss medical appointments, or ignore symptoms during this period of social restriction," he said.
"There are safe ways of getting all sorts of care, including consultations, vaccinations, prescriptions and more.
"You may be able to have a phone or video appointment, or even have your medicines delivered.
"Face-to-face appointments will still be recommended in some cases, but they can be done safely.
"There are rules about physical distancing (such as sitting a safe distance apart in the waiting room and during your consultation), and strict cleaning measures are also in place."
The Sutherland drive-through clinic is located at the Sutherland Multi-Purpose Community Centre, 123 Flora Street, and operates from 9am to 4pm, Monday to Friday (excluding public holidays).
No referral or appointment is required.
